In Vitro Plant Regeneration of Libyan Wild Plants: Edible Species (Arbutus pavarii) and Endanger Species [Haplophyllum tuberculatum (Forsk.) Juss]

Abstract :Protocol of micropropagation of Arbutus pavarii and Haplophyllum tuberculatum was successfully achieved. In vitro plant regeneration of Arbutus pavarii was attempted using single nodal on MS medium supplemented with different concentration of three various growth regulators indole-3-butyric acid (IBA), Kinetin (K) and N-Isopentenylaminopurine (2ip). Multiple shoots from single node were obtained on MS medium with the 0.5 mg/l 2ip. While, faster improving of shoots elongation was on MS contained 0.5 mg/l K. Explants obtained in micropropagation step were used for rooting step under several treatments. The best results were obtained when explants were sup-cultured on MS medium with 1 mg/l IBA. New plants were vigorous, of good quality and presented phenotypic characters similar to mother plants. Micropropagation of Haplophyllum tuberculatum was achieved from sterilized single nodal segments on MS medium supplemented with for different concentrations of 2,4-D for callus induction and other different hormones K and BA for multiplication of auxiliary branches and rooting . The highest results of the weight of callus were growing in MS medium containing 1 or 2 mg/l 2.4-D hormone with maltose. Whereas, the axillary soothing was significantly proliferated on MS medium supplemented 2 mg/ l -1 K.
