The interaction between a zinc(II) phthalocyanine compound bearing octakis phenoxyacetamide substituents and calf thymus DNA

Abstract : In this study, a zinc(II) phthalocyanine compound ( PcZn ) substituted with octakis phenoxyacetamide was synthesized and characterized. Calf thymus DNA (CT-DNA) was used to determine the DNA binding properties of the zinc(II) phthalocyanine compound. This interaction of CT-DNA with PcZn was investigated using electronic absorption spectra, emission spectroscopy, a thermal melting temperature study, cyclic voltammetry, and viscosity measurements in a Tris-HCl buffer solution at a pH of 7.2. The findings showed that zinc(II) phthalocyanine interacts with calf thymus DNA via partial intercalative binding mechanisms. In addition to the techniques above, a gel electrophoresis study was also carried out to verify the interaction of CT-DNA with the PcZn complex. The results demonstrated that the octakis phenoxyacetamide-substituted zinc(II) phthalocyanine complex binds to CT-DNA via a partial intercalative binding mode.Keywords : DNA binding, absorption spectroscopy, gel electrophoresis, thermal melting, phthalocyanine compound
