Determination of Contamination of Pseudomonas savastanoi pv. phaseolicola on Bean Seeds in Central Anatolia Region

Abstract :Halo blight disease of bean caused by Pseudomonas savastanoi pv. phaseolicola, is a bacterial agent transmitted by seeds and causing serious damages. Seeds of dry bean were collected from 12 provinces in Central Anatolia Region and economically bean growing areas based on size the sown field. Isolation and identification of pathogen was based growth on Nutrient Agar(NA), Modified sucrose peptone (MSP), King’s B, levan production, growth at 37 °C, oxidase reaction, pectolytic activity, arginine dehidrolase, utilizing of carbon sources, arbutin and gelatin hydrolysis, nitrate reduction, H2S production, hypersensitive reaction on tobacco (cv. White Burley) tests. Typical leaf symptoms were obtained on bean cultivar Dermason which was inoculated by a suspension 108 cfu/ml of P. s. pv. phaseolicola in the pathogenicity tests. Molecular detection of the agent was made by Bio-PCR method and amplification of phaseolotoxin gen. According to obtained data, the pathogen was detectected on 38 seed samples from totally collected 175 dry bean samples and 21,71% of the seed samples was found contaminated by the pathogen in Central Anatolia Region.Keywords : fasulye, tohum, Pseudomonas savastanoi pv. phaseolicola, Bio-PCR
