Morphological and Cytological Effects of Colchicine Treatments on ‘Narince’ (Vitis Viniıfera L.) Grape Variety

Abstract :Turkey is among the important countries in table grape production, consumption and international marketer. The table grape market is in an increasing trend and the devel-opment of the traditional varieties in this area and the breeding of new grape varieties are necessity. In this project stimulation of polyploidy with colchicine applications in \'Narince\' grape (Vitis vinifera L.) variety has been tested in vivo conditions. Single node cuttings ware rooted in greenhouse 1: 1 peat: perlite media in greenhouse, 4 colchicine dosages (0%, 0.25%, 0.5%, 0.75% and 1%) with glycerine 5% were applied by dropwise to the shoot tip with two different times (48 and 96 hours). After the application, some of the shoot tips were covered with aluminium foil and the other were left open, and the effects were assessed by the shoot tip viability rate, shoot length, stoma dimensions (length, width, area and density) and flow cytometry (FC) analysis. Although some changes in the morphological characteristics were observed, a significant change in the level of ploidy was not determined according to FC analysis. In \'Narince\' grape variety, the exact frequency of colchicine-induced polyploidy induction is less than 1/120 of the findings obtained from this study. Due to the detected morphological differences and variation in FC analysis, the material transferring it the open area has been monitoring to follow up the future developments.Keywords : Narince, Islah, Ploidi, Mutasyon, Flow sitometri
